Job Description

Key Responsibilities

  • Synthesis, purification, and characterization of antibody oligonucleotide conjugates (AOCs) and antibody drug conjugates (ADCs) on mg to gram scale under minimal supervision.
  • Design and execute engineered cysteine antibody conjugation workflows
  • Optimize reaction stoichiometry, linker chemistry, and reaction conditions to improve peptide-to-antibody ratios and product homogeneity.
  • Development, optimization, and validation of conjugation protocols specifically for antibody oligonucleotide conjugates.
  • Optimization and validation of formulation buffers for bioconjugates.
  • Application and refinement of automated techniques for AOC/ADC synthesis and purification.
  • Collaboration with cross-functional teams to advance AOC discovery programs from conceptualization to early development.
  • Develop and optimize peptide–antibody conjugation strategies
  • Optimize reaction stoichiometry, linker chemistry, and reaction conditions to improve peptide-to-antibody ratios and product homogeneity.
  • Investigate and mitigate sources of conjugation heterogeneity such as over-reduction, disulfide scrambling, hydrolysis, and aggregation.
  • Apply diverse conjugation chemistries including thiol-reactive linkers, maleimide coupling, NHS ester chemistry, and bioorthogonal click chemistry (e.g., azide–alkyne cycloaddition).
  • Characterize conjugates using analytical techniques including SEC-HPLC, LC-MS (intact/reduced), RP-HPLC, and SDS-PAGE.
  • Access external innovation to complement internal efforts

Qualifications

Qualifications

  • Bachelor’s Degree or equivalent education and typically, 12 years of experience, Master’s Degree or equivalent education and typically 10 years of experience, PhD and typically 4 years of experience.​
  • Expertise in the discovery, design, synthesis, and purification of Antibody Oligonucleotide Conjugates (AOC), including familiarity with associated chemistries and analytical methods.
  • Hands-on experience with relevant analytical techniques such as HIC-HPLC, SEC, IEX, RP-HPLC, and capillary electrophoresis techniques (e.g., CE-SDS, icIEF).
  • Proven problem-solving skills, motivation, enthusiasm, attention to detail, and the ability to thrive in a fast-paced, cross-disciplinary team environment.
  • Superior written and oral communication skills, with a strong record of collaboration and innovation.
  • Familiarity with biorthogonal conjugation techniques and next-generation site-specific conjugation platforms.
